This procedure traces building one recombinant DNA molecule, from cutting a foreign DNA fragment and a plasmid with the same restriction endonuclease through to a sealed, ligated vector.
Watch for
Ligation only succeeds where two sticky ends are complementary — a plasmid and a fragment cut with different, non-matching restriction endonucleases will not anneal correctly.
